Preparation of HSV-2 antigens:
HSV-2 is propagated in mammalian cells using a protocol optimized for maximum particle yield. At the appropriate time, infected cells are washed in saline then disrupted to release virus particles by freeze thaw and sonication.
Purification and presentation:
PBS lysate: Virus particles released from cells are partially purified by low speed centrifugation to remove cell debris. HSV-2 particles are then purified by sucrose density gradients. This process also includes a DNase step to remove cellular DNA and helps to achieve a high purity.
