APPLICATION
The protein produced with the ALiCE Cell-Free Protein Expression kit can be used for any application, including: Structural analysis experiments (including NMR and X-ray crystallography), Protein interaction studies, Enzyme assays, Membrane protein expression and analysis, Protein activity screening, Cryogenic electron microscopy
USAGE
The ALiCE kits were developed specifically for ease of use. Expression of your protein of interest follows a simple four-step procedure:
1. Clone your gene of interest into the ALiCE vector of your choosing
2. Add your plasmid DNA to a thawed vial of ALiCE lysate
3. Incubate at 25°C with aeration for 24-48 hours.
4. Collect your protein

The user should observe all applicable regulations for handling chemicals and recombinant DNA. Lab coat, safety glasses and gloves should be worn when handling the kit components. QUALITY CERTIFICATION / COMPLIANCE
High quality chemicals and materials have been used to manufacture the components of this kit. Each lot is carefully tested to ensure that all components meet the stated specifications (see Certificate of Analysis). Every batch of the ALiCE lysate undergoes rigorous quality control testing. Before release, the lysate must meet our stringent criteria of achieving 3 mg/mL eYFP following a 48 hour reaction period.

• Pipette filter tips (RNase-free)
• RNase-free water
• Plasmid purification kit for isolating RNase-free transfection-grade plasmid DNA. We recommend using the Macherey-Nagel™ NucleoBond™ Xtra kit.
• For reactions performed in tubes: Eppendorf ThermoMixer® series or similar heated table-top shaker with an equivalent shaking diameter of 3 mm.
• Recommended For reactions performed in microtiter plates: 96-well half area microplates and a shaker system with a humidity above 70%, run at 500 rpm with a shaking diameter of 12.5 mm. Additionally, we recommend using the Sandwich cover system “Duetz” from EnzyScreen BV (Heemstede, Netherlands) together with their universal clamp holder system. Please follow the recommendations for the microtiter plate setup if humidity control is missing.

WHAT IS ALiCE®?
ALiCE® is a plant-based cell-free lysate derived from Nicotiana. tabacum c.v. BY-2 root cells. ALiCE® stands for Almost Living Cell-Free Expression. The lysate is prepared by removing the cell wall and other cell components that are not necessary for protein production, leaving only the the protein production machinery and mitochondria which power the reaction. During our lysate finalization steps, the additional components necessary for protein production (amino acids, co-factors, etc.) are added to produce something that functions just like a cell, but without the constraints of a living system - all that's needed is the DNA encoding the protein of interest!
