Storage Temperature
Remove the flask from the shipping pouch. Do not remove the seal or the Transport Medium. Place the flask in a 37ºC incubator. Stand the flask on its end to expose maximum surface area to the warm, humidified, circulating atmosphere. Do not place the flask flat. The steel shelf will give up heat to the cell-bearing bottom surface of the flask much too fast, sandwiching the cells between a heat source (the shelf) and a heat sink (the Transport Medium). This is too much ∆T (change in temperature) for the cells to accommodate without significant damage to the cytoskeleton. Two to three hours later, during which time the feeding medium has been warmed to 37ºC in a water bath, aspirate (and discard) the Transport Medium and feed with 5-7 ml Complete Classic Medium (4Z0-500) supplemented with CultureBoost™. Transport Medium is neither intended nor suitable for maintaining cells in the incubator.
