Tetracycline: a Broad Spectrum Polyketide Antibiotic and a Selective Reagent Genetically Engineered Cells
Tetracycline is a polyketide antibiotic isolated from Streptomyces genus. Tetracycline has broad activity against Gram-negative bacteria and Gram-positive bacteria, with a few exceptions, such as Pseudomonas aeruginosa and Proteus species. Tetracycline inhibits protein synthesis in susceptible organisms by binding to the A-site on the 50S subunit of a bacterial ribosome. However, tetracycline-resistant bacterial strains appeared soon after its introduction. Tetracycline is used in molecular biology as a selective reagent most commonly to isolate bacteria coupled to a gene coding for tetracycline resistance, which is one of the most frequently used selection markers for obtaining transgenic cells. Recently, tetracycline is used for tetracycline-controlled transcriptional activation systems in cells. (The product is for research purpose only.)
