ADAM12 (A disintegrin and metalloprotease domain 12; also meltrin-α) is a 120kD type I transmembrane glycoprotein member of the metzincin family of proteases. It is synthesized as a 909 amino acid (aa) preproprecursor that contains a 25kD proregion (aa 29-207) and a 92kD mature molecule. Proteolytic cleavage in the Golgi removes the propeptide prior to cell-surface expression. A 68kD soluble splice form exists in pregnancy where 36 aa replace the C-terminal 205 aa (705-909).
