Calpain 1 (mu-calpain), a calcium-dependent cysteine C2 endopeptidase, is involved in myofibrillar protein degradation. In skeletal muscles, activation of Calpain I results in degradation of skeletal structural proteins, improving meat tenderness. Calpains I is inhibited by the endogenous inhibitor calpastatin and by leupeptin. Calpastatin is inactivated in postmortem skeletal muscle. Activity and expression of calpains in peripheral lymphoid organs may play an important role in T cell migration.
